Methods: The mRNAs from the cell lines SW480, SW620 and SW480/M5 of different metastatic abilitywere labeled with biotin respectively, and were hybridized with expression profile microarray. The bioinformatic method was used topreliminarily analyze the gene FMNL2, and the expression of FMNL2 in the cell lines SW480, SW620 and SW480/M5 with differentability of metastasis was detected using fluorescent quantitation PCR. Results: A total of 422 differentially expressed genes were dis-covered by the expression profile microarray,. Bioinformatic analysis was conducted to explore the structure, chromosomal localization,physico-chemical property of the encoded protein, subcellular localization and functional domain of the upregulated gene FMNL2. Thesystematic evolution analysis of the similar proteins of FMNL2 was also performed. It basically confirmed a conservative nucleoproteinencoded by the gene in the high expression and evolution of the embryo and tumor cells. The real time quantitative PCR detection ofFMNL2 gene found that the gene expression was 17.8 and 13.9 times higher in the SW620 and SW480/M5 cells than in the SW480cells. Conclusion: Bioinformatic analysis indicates that FMNL2 is a tumor associated protein which may involve embryogenesis and tu-mor metastasis.Keywords cDNA gene; Gene expression profile microarray; Colorectal carcinoma; Bioinformatics; FMNL2
